OD600 Calculator Measurement Principles & Turbidity Physics
Optical density measured at 600 nm (OD600) is the standard method used in molecular biology laboratories to estimate microbial cell density in liquid suspension. Unlike molecular absorption by dyes or nucleotides, OD600 measures light scattering (turbidity) caused by bacterial and fungal cells rather than true chemical absorption.
| Model Organism | Cells/mL at OD 1.0 | Dry Cell Weight | Wet Biomass | Common Stage |
|---|---|---|---|---|
| Escherichia coli | 8.0 × 10⁸ cells/mL | 0.39 g/L | ~1.7 g/L | IPTG induction: OD 0.6 – 0.8 |
| Saccharomyces cerevisiae | 3.0 × 10⁷ cells/mL | 0.50 g/L | ~2.2 g/L | Log phase: OD 0.5 – 1.0; Yeast Transformation |
| Bacillus subtilis | 5.0 × 10⁸ cells/mL | 0.36 g/L | ~1.5 g/L | Competence prep: OD 0.8 – 1.0 |
| Pichia pastoris | 5.0 × 10⁷ cells/mL | 0.65 g/L | ~3.0 g/L | Methanol induction; high-density fed-batch |
Spectrophotometer Linearity & Dilution Best Practices
The linear OD600 range depends on the instrument, path length, organism, and culture conditions. OD 0.1–0.8 is a practical working guide; verify linearity for your sample by measuring a dilution.
Above about OD600 0.8, readings may depart from linearity; the deviation depends on the instrument and sample. Dilute into roughly 0.1–0.6 and check whether dilution-corrected OD agrees across dilution factors.
OD > 0.8: Multiple Scattering
Photons scattered by one bacterium are re-scattered back into the detector path by another bacterium, causing the instrument to underestimate true cell density.
Correct Protocol: Serial Dilution
Dilute dense bacterial suspensions 1:5 or 1:10 with blank growth media so the measured OD sits between 0.1 and 0.6. Multiply the reading by the dilution factor.
Worked Lab Example: Shake Flask Induction
Scenario: Inducing E. coli BL21(DE3) in a 1 Liter Shake Flask
You take a 100 µL aliquot of your culture, dilute it into 900 µL LB medium (10-fold dilution, DF = 10), and measure OD600 = 0.28 on the spectrophotometer.
- 1. Corrected True OD = 0.28 × 10 = 2.80
- 2. Cell Concentration = 2.80 × 8×10⁸ = 2.24 × 10⁹ cells/mL
- 3. Total Bacterial Count in 1 L = 2.24 × 10⁹ cells/mL × 1000 mL = 2.24 × 10¹² cells
- 4. Dry Cell Biomass = 2.80 × 0.39 g/L = 1.092 g DCW
Frequently Asked Questions (FAQ)
Why is 1.0 OD600 equal to 8 × 10⁸ cells/mL for E. coli?
For a standard 1 cm light path spectrophotometer, 1.0 OD600 in LB medium corresponds to approximately 8 × 10⁸ E. coli cells/mL (empirical range 5×10⁸ to 1×10⁹ depending on optical geometry and strain size).
Why must I dilute dense cultures with OD600 > 0.8?
Above ~0.8 OD, multiple light scattering occurs where scattered photons re-enter the detector path, violating the Beer-Lambert linear relationship and leading to severe underestimation of true cell density.
Does OD600 measure viable living cells or total biomass?
OD600 measures light scattering caused by all particles in suspension, including dead cells, intact debris, and insoluble aggregates. For live-cell viability counts, plate serial dilutions for Colony Forming Units (CFU).
Can I use water to blank the spectrophotometer for OD600?
No. Always blank with the sterile growth medium used for culturing (e.g. LB, YPD, Terrific Broth). Media components like yeast extract absorb significantly at 600 nm.
